FDA approvalThe FDA approved daraxonrasib (RMC-6236), the first RAS-targeted therapy for previously treated metastatic pancreatic adenocarcinoma. In the 500-patient RASolute 302 trial, oral daraxonrasib doubled median overall survival versus chemotherapy (13.2 vs. 6.7 months; HR 0.40), with median progression-free survival of 7.2 vs. 3.6 months (HR 0.49).
Phase 3 trialDaraxonrasib, an oral RAS(ON) multiselective inhibitor, doubled overall survival vs. chemotherapy in previously treated metastatic pancreatic cancer (13.2 vs. 6.6 months; HR 0.40). Led by Dr. Eileen M. O’Reilly, Dr. Wungki Park’s mentor at MSK, and the RASolute 302 Trial Investigators.
The first clinical study of daraxonrasib in 168 patients with previously treated RAS-mutated pancreatic cancer reported a 35% objective response rate, median progression-free survival of 8.5 months, and median overall survival of 13.1 months in the second-line RAS G12 cohort receiving 300 mg. Dr. Wungki Park was a co-author.
Led by first author Dr. Daeseong Kim, this clinicogenomic study identified PI3K-pathway activation as a molecular determinant of outcome with gemcitabine, cisplatin, and nab-paclitaxel in advanced biliary tract cancer, supporting biomarker-guided treatment selection and the lab’s expanding GRIT initiative.
